SLC44A4 (solute carrier family 44 member A4) is a gene encoding a protein that primarily functions as the thiamine pyrophosphate transporter (TPPT) in humans. This protein is responsible for the absorption of thiamine pyrophosphate (TPP), a major form of vitamin B1, generated by gut microbiota in the large intestine. The SLC44A4 gene is highly expressed in the colon and is localized to the apical membrane of colonic cells, enabling efficient uptake of TPP. This transporter operates in a sodium-independent manner and is crucial for maintaining thiamine homeostasis, which is vital for various cellular processes including carbohydrate metabolism and neural function. Disruption or mutations in SLC44A4 can impact the body's ability to absorb thiamine, leading to deficiencies that may result in metabolic and neurological disorders.
