Easily and quickly quantify inhibitors of SARS-CoV-2 3C-like protease (3CLpro) and papain-like protease (PLpro) using the EnzChek 3CLpro and PLpro Assay kits. These kits can be used to screen and profile inhibitors of 3CLpro and PLpro and include all reagents for the determination of protease activity, including a positive control inhibitor. 3CLpro and PLpro activity can be directly correlated using a 96-well plate and microplate reader.

The EnzChek 3C-like Protease (3CLpro) and Papain-like Protease (PLpro) Assay kits are quick, easy, and direct assays for detecting and quantifying SARS-CoV-2 3-chymotrypsin-like protease (3CLpro) and SARS-CoV-2 papain-like protease (PLPro) activity. 3CLpro, also known as main protease (Mpro), and PLpro are crucial enzymes for processing viral polyproteins, and due to their role in the SARS-CoV-2 virus life cycle have been identified as a promising drug target for blocking viral replication. The homogeneous EnzChek 3CLpro and PLpro Assay kits are ideal for easily screening and quantifying inhibitors of 3CLpro and PLpro.

The EnzChek 3C-like 3CLpro and PLpro Assay kits contain a fluorogenic protease-specific substrate, SARS-CoV-2 3CLpro or PLpro, positive control inhibitor, and assay buffer. Simply add your inhibitor of choice. Protease-catalyzed hydrolysis releases the fluorescently labeled peptide, and protease activity can be directly quantified from the change in fluorescence signal using a microplate reader (ex: 360 nm, em: 460 nm).
