Urabrelimab, also known as SRF231, is an investigational human monoclonal antibody targeting the CD47 protein, a cell surface antigen involved in immune evasion by cancer cells. The CD47 gene encodes a transmembrane glycoprotein broadly expressed on the surface of various cell types. Functionally, CD47 serves as a marker of self by binding to the Signal-regulatory protein alpha (SIRP alpha) receptor on myeloid cells, thereby preventing macrophage-mediated phagocytosis. This interaction is crucial in maintaining homeostasis by inhibiting the immune system from attacking healthy cells. However, CD47 is significantly overexpressed on the surface of many tumor cells, providing a mechanism for these cells to evade the immune system.
