Preparation: Produced from sera of goats immunized with highly pure Recombinant Human TWEAK. Anti-Human TWEAK-specific antibody was purified by affinity chromatography employing an immobilized Human TWEAK matrix. Sandwich ELISA: To detect Human TWEAK by sandwich ELISA (using 100 μL/well antibody solution) a concentration of 0.5-2.0 μg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human TWEAK (500-P137GBt) as a detection antibody, allows the detection of at least 0.2-0.4 ng/well of Recombinant Human TWEAK. Western Blot: To detect Human TWEAK by Western Blot analysis this antibody can be used at a concentration of 0.1- 0.2 μg/mL. Used in conjunction with compatible secondary reagents the detection limit for Recombinant Human TWEAK is 1.5-3.0 ng/lane, under either reducing or or non-reducing conditions.

TNF-related weak inducer of apoptosis (TWEAK) is a member of the tumor necrosis factor superfamily (TNFSF) of structurally related cytokines. Like most other members of this family, TWEAK is a cell surface-associated type II transmembrane protein although a smaller, biologically active form can also be generated by cleavage near the cell membrane. TWEAK has multiple biological activities, including stimulation of cell growth and angiogenesis, induction of inflammatory cytokines, in addition to stimulation of apoptosis. The TWEAK signal transduction pathway has not been well established but it appears to signal via TweakR/Fn14 in a manner similar to that described for other TNFSF members that bind receptors lacking death domains.
