Macherey-Nagel™ Elution Buffer AE
Description
NucleoSpin™ Plasmid kits feature a protocol that concludes with an elution step. Plasmid DNA Elution Buffer AE is a 5mM Tris/HCl solution with a pH of 8.5 for extracting pure plasmid DNA from the matrix of the NucleoSpin™ column under low ionic strength conditions.
Supplemental Elution Buffer AE is necessary when adapting the buffer volume to achieve a higher yield, which may be required for some downstream applications. NucleoSpin™ Plasmid kits include enough Elution Buffer AE to accommodate the standard protocol (typical recovery: 70–90%).
- Higher yield in general, esp. for larger constructs: Heat buffer, use up to 2x the buffer, incubate longer
- High yield: Perform two elution steps, which uses 2x the elution buffer, for a recovery of 90–100%
- High concentration: Use 60% volume of elution buffer for ∼130% concentration (max. yield: 80%)
- High yield and high concentration: Perform a longer incubation using 50% volume of elution buffer, then apply a standard elution step (yield: 85–100%, at a high concentration)
