Conditions for 100% Activity:

• 1X OPTIZYME Buffer 2: 10mM Tris-HCl (pH 7.5 at 37°C), 10mM MgCl₂, 50mM NaCl and 0.1mg/ml BSA
• Incubate at 55°C

Enzyme Activity in OPTIZYME buffers:

Ligation and Re-cleavage:

More than 95% of the DNA fragments can be ligated and re-cut after 10-fold over-digestion with BclI.

Notes:

• Incubation at 37°C results in 50% activity.
• Greater than 15-fold overdigestion with BclI may result in star activity.
• Assayed using lambda DNA (dam-)
• BclI is blocked by dam methylation. To avoid dam methylation, use a dam-, dcm- strain.
