NucleoSpin™ 96 Tissue is designed for the rapid simultaneous preparation of highly pure genomic DNA from animal and human tissue, eg. mouse and rat tails.
From a 1.2cm mouse tail tip section (age of mice 4 to 6 weeks), up to 30μg to 40μg of pure genomic DNA can be prepared (typical yields 15μg to 25μg). The obtained DNA can be used directly for PCR*, Southern blotting or any kind of enzymatic reaction.NucleoSpin™ Tissue binding plates can be used under vacuum or centrifugation in a centrifuge with deep buckets allowing processing of NucleoSpin™ 96 well plates stacked on 96 well blocks. Up to 192 samples (two plates) can be processed in less than one hour (after lysis). With the NucleoSpin™ 96 Tissue method, lysis is achieved by incubation of the samples in a solution containing detergents and proteinase K at 56°C. Appropriate conditions for binding of DNA to the silica membranes of the NucleoSpin™ Tissue binding plate are created by addition of large amounts of chaotropic ions plus ethanol to the lysate. The binding process is reversible and specific to nucleic acids. Contaminants are removed by washing with two different buffers. Pure genomic DNA is finally eluted under low ionic strength conditions in a slightly alkaline elution buffer.Kit components: NucleoSpin™ Tissue binding plates, buffers, proteinase K, wash plates, square-well blocks, round-well blocks, racks with tube strips, cap strips, self-adhesing foil.
