Description

NucleoSpin™ 8/96 Plant is designed for the isolation of genomic DNA from plant tissue (e.g., Arabidopsis, tobacco or maize). Depending on the individual sample (20mg to 100mg) typical yields are in the range from 5 to 30μg DNA. For RNA-rich samples, it is recommended to add RNase to the lysis buffer or perform a subsequent RNase treatment. The general isolation procedure ensures lysis of plant material with a CTAB lysis buffer developed especially for plant samples. CTAB is removed completely in the subsequent washing steps and thus does not interfere with downstream PCR*. The binding process is reversible and specific for nucleic acids. Contaminations are removed by washing. Pure genomic DNA is finally eluted under low ionic strength conditions in a slightly alkaline elution buffer. The obtained DNA can be used directly for PCR*, Southern blotting or any kind of enzymatic reaction. NucleoSpin™ 8/96 Plant can be used in a centrifuge with deep buckets allowing processing of NucleoSpin™ 8 well strips/96 well plates stacked on 96 well blocks. NucleoSpin™ 8/96 can also be used under vacuum, manually in the NucleoVac 96, or automated on many common liquid handling instruments. Up to 192 samples (two full 96 well plates) can be processed in 2 to 3hrs depending on equipment used for pipetting and homogenisation.
Required hardware
Centrifugation
For centrifuge processing of NucleoSpin 8/96 plant kits a microtiterplate centrifuge is required which is able to accommodate the NucleoSpin Plant strips/plate stacked on a round or square well block or rack with tube strips and reaches accelerations of 5, 600xg to 6, 000xg (bucket height: 85mm). For further information on suitable centrifuges please contact our Technical Service team. For centrifuge processing of the 8 well Strips, the reusable Starter Set C, containing column holders C, dummy strips, MN square well blocks, and tube strips, is required.
Vacuum processing
The NucleoSpin 8/96 plant kits can be used manually with the NucleoVac 96 vacuum manifold. Additionally, a suitable centrifuge for sample preparation steps may be required. For vacuum processing of the 8 well strips the reusable Starter Set A, containing column holders A and dummy strips is required. For automation on laboratory platforms with standard 96 well plate vacuum chambers the use of the Starter Set A is also required.
