Mupadolimab is a humanized monoclonal antibody that targets CD73, an enzyme expressed on various immune cells, including B cells and T cells. CD73 plays crucial roles in processes such as immunomodulation and adenosine production. CD73 (also known as ecto-5'-nucleotidase) is encoded by the NT5E gene located on chromosome 6q14-q21. It is a glycosylphosphatidylinositol (GPI)-anchored membrane protein that catalyzes the dephosphorylation of extracellular AMP to adenosine and inorganic phosphate. Functionally, CD73 plays critical roles in various physiological and pathological processes. It modulates immune response by generating adenosine, which has immunosuppressive effects, thus aiding tumor cells in evading the immune system. CD73 is implicated in cancer cell proliferation, migration, invasion, and tumor angiogenesis, making it a significant target for cancer immunotherapy. Its expression is also associated with memory T-cell function, suggesting its pivotal role in adaptive immunity. Moreover, mutations in NT5E are linked to a rare genetic disorder known as arterial calcifications due to deficiency of CD73.
