IL-5 is a cytokine that acts as a growth and differentiation factor for both B cells and eosinophils. IL-5 cytokine is a main regulator of eosinopoiesis, eosinophil maturation and activation. The elevated production of this cytokine is reported to be related to asthma or hypereosinophilic syndromes. The receptor of IL-5 cytokine is a heterodimer, whose beta subunit is shared with the receptors for interleukine 3 (IL3) and colony stimulating factor 2 (CSF2/GM-CSF). IL-5, together with those for interleukin 4 (IL4), interleukin 13 (IL13), and CSF2, form a cytokine gene cluster on chromosome 5. IL-5, IL4, and IL13 are found to be regulated coordinately by long-range regulatory elements spread over 120 kilobases on chromosome 5q31. Description: Recombinant mouse IL-5 is a disulfide-linked homodimer, containing two 113 amino acid peptides; these resolve as 32-34 kDa bands in SDS-PAGE. IL-5 is produced by T cells and has been known as eosinophil-differentiating factor (EDF), B cell growth factor II (BCGFII), and T cell-replacing factor (TRF). IL-5 induces eosinophil differentiation and promotes eosinophil survival and activation. In mice, IL-5 has been shown to stimulate B cell proliferation and antibody production. Applications Reported: Recombinant mouse IL-5 is biologically active. The intended use of the Recombinant Mouse IL-5 Single-Use ELISA RSG Standard is as a standard for mouse IL-5 Ready-SET-Go! ELISA and mouse IL-5 matched antibody pair ELISA. Typically the standard is run in a series of 7 doubling dilutions from the ′top′ standard described below. Instructions for use: For Ready-SET-Go! ELISA Set (TMB substrate). Dilution: Add 5uL of standard solution to 10 mL of assay diluent to prepare the top standard solution (500 pg/mL).For matched antibody pair ELISA (ABTS or Super AquaBlue substrate). Dilution: Add 10uL of standard solution to 10 mL of assay diluent to prepare the top standard solution (1000 pg/mL). Note: These are single-use vials. Use one time and discard. Dilute as suggested above, not difficult-to-measure fractions of the suggested volumes (e.g., not 0.5uL in 1.0 mL) or recovery may not be accurate. Use within 6 hours of dilution to top standard concentration. Applications Tested: This cytokine standard has been calibrated, and subsequently tested by sandwich ELISA using the mouse IL-5 ELISA pair (TRFK5/TRFK4).
