Latozinemab, also known as AL001, is a biosimilar human monoclonal immunoglobulin G1 antibody targeting the sortilin receptor, SORT1. Its primary function is to elevate progranulin (PGRN) levels by blocking sortilin, which is implicated in the degradation pathway of PGRN. The SORT1 gene encodes Sortilin 1, a membrane receptor primarily located on the membranes of the endoplasmic reticulum and lysosomes. It plays a critical role in protein transport, particularly directing proteins to lysosomes. SORT1 has been identified as a high-affinity neuronal receptor of Progranulin (GRN), mediating its endocytosis and guiding it toward the endolysosomal pathway, which results in reduced extracellular GRN levels. In Sort1 knockout mice, GRN levels are increased in the brain and serum, implicating SORT1's role in regulating GRN homeostasis. Clinically, SORT1 expression has shown a significant correlation with coronary artery disease (CAD) and its severity, serving as a potential diagnostic marker. Studies also indicate its involvement in ovarian cancer progression by enhancing cell proliferation, migration, and invasion. The SORT1 gene is located at the cardiovascular risk locus 1p13.3, near genes CELSR2 and PSRC1, and it is implicated in modulating cardiovascular risk, although its specific role remains to be fully elucidated. Additionally, the miR-182/SORT1 axis is noted for regulating vascular smooth muscle cell behaviors, providing further insights into its regulatory functions in vascular biology.
