MHC Class II (I-Ap) is a protein complex found on the surface of cells in vertebrates. Its main function is to present peptides derived from extracellular proteins to T cells for immune recognition. MHC Class II (I-Ap) is composed of an alpha and beta chain, both of which contribute to the peptide-binding groove. MHC Class II (I-Ap) is primarily restricted to antigen-presenting cells, including dendritic cells, macrophages, and B cells. These cells are responsible for capturing and processing antigens from the extracellular environment. Once the antigens are internalized, they are broken down into smaller peptides, which then bind to MHC Class II molecules within the endosomal compartments of these cells. MHC Class II (I-Ap) consists of an extracellular domain, a transmembrane domain, and a cytoplasmic tail. The extracellular domain is responsible for interacting with the antigenic peptides, while the transmembrane domain anchors the MHC Class II molecule to the cell membrane. The cytoplasmic tail plays a role in intracellular signaling events. HLA and MHC antibodies play a significant role in Immunopeptidomics, facilitating the identification and characterization of neoantigens through high-performance liquid chromatography coupled to tandem Mass Spectrometry.
