• Recognizes human HDAC1
• HDAC1 (histone deacetylases) are a class of enzymes that remove acetyl groups from lysine residues leading to formation of condensed and transcriptionally silenced chromatin
  • Protein encoded by gene belongs to histone deacetylase/acuc/apha family
    • Component of histone deacetylase complex, responsible for gene expression silencing
  • Plays role in control of cell proliferation and differentiation by interacting with RB, p53 and other transcription factors
  • HADC inhibition causes apoptosis in tumor cells and HDAC inhibitors may be developed as anti-cancer agents

Process:

• Antibody specific for HDAC1 pre-coated onto microwells
• HDAC1 protein in samples captured by the coated antibody after incubation
• After extensive washing, another antibody specific for HDAC1 is added to detect captured HDAC1 protein
• For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
• Solution containing sulfuric acid used to stop color development and color intensity
  • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
