• Recognizes human haptoglobin
• Haptoglobin (HP, zonulin) is a plasma glycoprotein belonging to peptidase S1 family
  • Acute-phase response protein, serum Hp is mainly synthesized by hepatocytes and distributed in circulation
  • Main physiological function of serum Hp is binding free haemoglobin released from red blood cell lysis and protecting tissues and vessels from oxidative damage
  • Level Hp useful biomarker for some diseases diagnosis (Hepatocellular carcinoma, pancreatic cancer, Type 2 diabetes mellitus, diabetes kidney disease)

Process:

• Antibody specific for haptoglobin pre-coated onto microwells
• Haptoglobin protein in samples captured by the coated antibody after incubation
• After extensive washing, another antibody of biotinylated specific for human haptoglobin is added to detect captured human haptoglobin protein
• For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
• Solution containing sulfuric acid used to stop color development and color intensity
  • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
