Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous CXCL16

• Recognizes human CXCL16
• CXC chemokine ligand 16 (CXCL16) is a CXC soluble chemokine, adhesion molecule and cell surface scavenger receptor
  • Regulates inflammation, tissue injury and fibrosis
  • CXCL16 produced by several inflammatory cells (macrophages, dendritic cells, smooth muscle cells, lymphocytes)
  • Expressed in various cancers (pancreatic, prostate, breast, colorectal, non small cell lung cancer)
  • Proposed as pathogenic mediator in inflammatory diseases (rheumatoid arthritis, hepatitis, atherosclerosis)

Process:

• Antibody specific for CXCL16 in samples pre-coated onto microwells
• CXCL16 protein in samples captured by the coated antibody after incubation
• After extensive washing, another antibody of biotinylated specific for CXCL16 is added to detect captured CXCL16 protein
• For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
• Solution containing sulfuric acid used to stop color development and color intensity
  • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
