• Recognizes human CX3CL1
• CXCL1 (fractalkine, FKN) is a chemokine with a transmembrane domain and chemokine domain on top of a long mucin like stalk
  • Can exist in membrane-bound form or shet soluble form, after extracellular proteolysis at membrane-proximal dibasic cleavage site
  • Soluble CX3CL1 has potent chemoattractant activity for T cells, monocytes and cell-surface-bound protein, induced by primary proinflammatory signals in activated primary endothelial cells
    • Promotes strong adhesion of leukocytes
  • Presence of CX3CL1 and receptor CX3CR1 in lymphocytes and enhanced cellular expression during inflammatory conditions suggest CX3CL1-CX3CR1 duo participates in immune-related inflammatory diseases

Process:

• Antibody specific for CX3CL1 pre-coated onto microwells
• CX3CL1 protein in samples captured by the coated antibody after incubation
• After extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for CX3CL1 is added to detect captured CX3CL1 protein
• For signal development, followed by Tetramethyl-benzidine (TMB) reagent
• Solution containing sulfuric acid used to stop color development and color intensity
  • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
