• Recognizes human CD22
• CD22 (Siglec-2, BL-CAM) is a 130-140 kDa, B-cell restricted, Type I transmembrane glycoprotein belonging to immunoglobulin gene superfamily
  • Expression of CD22 developmentally regulated
  • Expressed at low levels in cytoplasm of pro-B and pre-B cells, present on cell surface only at mature stages of B-cell differentiation
  • CD22 inhibitory receptor for BCR signalling, preferntially binds to alpha-2,6-linked sialic acid and mediates B-cell B-cell interactions
  • Plays role in activation and differentiation of B-cell
  • Soluble form of CD22 found in human serum, useful as tumor marker for hairy cell leukemia

Process:

• Antibody specific for CD22 pre-coated onto microwells
• CD22 protein in samples captured by the coated antibody after incubation
• After extensive washing, another antibody of biotinylated specific for human CD22 is added to detect captured human CD22 protein
• For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
• Solution containing sulfuric acid used to stop color development and color intensity
  • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
