Description

Description

Description: The N2B2 monoclonal antibody reacts with mouse TNF-related apoptosis-inducing ligand (TRAIL), a member of the TNF superfamily. TRAIL is not detected on the surface of freshly isolated T, B, or NK cells, but can be induced preferentially on CD3- NK1.1+ NK cells after stimulation with IL-2 or IL-15. N2B2 inhibits IL-2- or IL-15-activated NK cell cytotoxicity against mouse fibrosarcoma L929 target cells. Applications Reported: This N2B2 antibody has been reported for use in flow cytometric analysis. Applications Tested: This N2B2 antibody has been tested by flow cytometric analysis of mouse CD253 transfected cells and can be used at less than or equal to 0.125 μg per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. Excitation: 633-647 nm; Emission: 660 nm; Laser: Red Laser. Filtration: 0.2 μm post-manufacturing filtered.

Apoptosis, or programmed cell death, occurs during normal cellular differentiation and development of multicellular organisms. Apoptosis is induced by certain cytokines including TNF and Fas ligand in the TNF family through their death domain containing receptors, TNFR1 and Fas. A novel member in the TNF family was recently identified and designated TRAIL (for TNF-related apoptosis-inducing ligand) and Apo-2L (for Apo-2 ligand) (6,7). TRAIL is a type II membrane protein and expressed in a variety of human tissues. Two novel death domain containing receptors DR4 and DR5 have been identified as the receptor for TRAIL3-6. Like TNF and Fas ligand, TRAIL induces apoptosis and NF-kB activation in many tissues and cells.
