The monoclonal antibody OX41 reacts with rat CD172a, which is also known as signal-regulatory protein alpha (SIRPa). SIRP family members are transmembrane glycoproteins with 3 Ig-like extracellular domains and a cytoplasmic tail containing 2 immunocreceptor tyrosine-based inhibition motifs. CD172a is expressed by monocytes, macrophages, granulocytes, dendritic cells, and neurons. SIRPa association with CD47 inhibits phagocytosis of CD47 presenting cells. Studies suggest that SIRPa ligation prevents phagocytosis by inactivating paxillin and cofilin, proteins involved in cytoskeleton remodeling.
