Description: The MBM7C7 monoclonal antibody reacts with human and mouse BATF (B cell activating transcription factor). BATF is a 14 kDa member of the AP-1 family of transcription factors and is involved in the repression of other AP-1 family members. BATF is upregulated in activated T cells when compared to naive T and B cells and has been shown to be critical for Th17, but not Th1 or Th2 differentiation. In Th17, BATF interacts with the promoters of IL17, IL17F, IL21 and IL22. BATF has also been shown to be involved in CD8 T cell exhaustion in HIV infection. Applications Reported: This MBM7C7 antibody has been reported for use in intracellular staining followed by flow cytometric analysis. Applications Tested: This MBM7C7 antibody has been tested by intracellular staining and flow cytometric analysis using the Foxp3/Transcription Factor Buffer Set (Product # 00-5523-00) and protocol. Please see BestProtocols® Section (Staining intracellular Antigens for Flow Cytometry) for staining protocol (refer to Protocol B: One-step protocol for intracellular (nuclear) proteins). This can be used at less than or equal to 0.5 μg per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. BATF is a nuclear basic leucine zipper protein that belongs to the AP-1/ATF superfamily of transcription factors. The leucine zipper of this protein mediates dimerization with members of the Jun family of proteins. This protein is thought to be a negative regulator of AP-1/ATF transcriptional events and blocks cellular transformation by Ras and Fos. BATF also is required for the differentiation of IL-17-producing T helper (TH17) cells, a CD4+ subset of T cells that coordinates the inflammatory response in host defense. In both T cells and B cells, BATF is required for the appropriate regulation of the class-switch recombination.
