Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous IL6

• Recognnizes human IL6
• IL6 is an interleukin that acts as pro-inflammatory and anti-inflammatory cytokine
  • Plays essential role in final differentiation of B-cells into Ig-secreting cells involved in lumphocyte and monocyte differentiation
  • Induces myeloma and plasmacytoma growth and induces nerve cell differentiation

Process:

• Antibody specific for IL6 pre-coated onto microwells
• IL6 protein in samples captured by coated antibody after incubation
• After extensive washing, another antibody specific for IL6 is added to detect captured IL6 protein
• For signal development, Sterptravidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
• Solution containing sulfuric acid used to stop color development and color intensity
  • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
