Fungal cell membranes contain a sterol component called ergosterol, which plays a role in maintaining the integrity and structure of the membrane. Amphotericin B has a high affinity for ergosterol, and it forms complexes with this sterol in the fungal cell membrane.
Once bound to ergosterol, amphotericin B forms channels or pores in the fungal cell membrane. These channels lead to an increase in the permeability of the membrane to ions, particularly potassium, and other molecules. The disruption of the fungal cell membrane compromises its integrity and disrupts the normal functions of the membrane.
The extensive damage to the fungal cell membrane and the loss of cellular components ultimately result in the death of the fungal cell. The antifungal activity of amphotericin B is fungicidal, meaning it directly kills the fungal cells rather than inhibiting their growth.
