Applications
3,3′-Diaminobenzidine is used to detect fingerprints in blood. 3,3′-Diaminobenzidine is used in immunohistology and immunoblotting as a precipitating substrate for the localization of peroxidase activity. Slides stained with DAB may be coverslipped in the traditional manner and stored for future reference. Many enhancement methods have been reported for DAB involving different buffer conditions, the addition of metal ions and post-treatment applications. DAB has been used in immunohistochemical staining of nucleic acids and proteins. Application of the silver-gold intensified 3,3′ -diaminobenzidine chromogen to the light and electron microscopic detection of the luteinizing hormone-releasing hormone system of the rat brain. 3, 3′-diaminobenzidine (DAB)-H2O2-horseradish peroxidase (HRP) has been presented and used for the sensitive detection of carcinoembryonic antigen (CEA) in human serum.

Each tablet contains 5mg of DAB, each tablet weighs 240mg. One tablet will yield 10 ml of substrate solution, enough to effectively detect 50cm² of membrane or about 20 slides. Insoluble product. Reaction Condition: Dissolve one 5mg tablet in 10 ml of 50mM Tris, 0.1M NaCl, pH 7.6 and 0.01% hydrogen peroxide. The solution may be filtered through a 0.2µm filter immediately prior to use if necessary. The presence of nickel or cobalt increases sensitivity.
