Endotoxin: 0.40 EU/mg Application Notes: IHC (frozen) - tissue sections can be fixed in acetone or 2% paraformaldehyde. Western blot - use non-reducing conditions. Flow - this antibody weakly stains alveolar macrophages by recognizing the extracellular domain of MARCO, with transfected COS cells used as a positive control. Immunoprecipitation - will immunoprecipitate MARCO as a 60 kDa and 50 kDa protein from transfected COS lysates. Functional Assays - blocks human alveolar macrophages by recognizing the extracellular doamin of MARCO. Recommended positive control: Human alveolar macrophages; Negative control: All other human cells.

MARCO, also known as SR-A6 or SCARA2, is a macrophage surface receptor and a member of the class A scavenger receptor family. As a part of the innate antimicrobial immune system, MARCO can bind both Gram-negative and Gram-positive bacteria via an extracellular, C-terminal, scavenger receptor cysteine-rich (SRCR) domain. In addition to short cytoplasmic and transmembrane domains, there is an extracellular spacer domain and a long, extracellular collagenous domain. The protein may form a trimeric molecule by the association of the collagenous domains of three identical polypeptide chains. Due to its expression in tumor infiltrating macrophages, and its role in inhibiting immune responses within tumor environment, MARCO has been indicated as a potential target for cancer immunotherapy.
