Description

High throughput purification of small and large constructs. Rapid Mini preparation of plasmid DNA in proven 96‐well format.

The NucleoSpin™ Multi-96 Plus Plasmid kit allows the fast purification of high copy-number plasmid DNA from E. coli in the 96 well format using a vacuum manifold or a benchtop centrifuge.

Sequencing grade plasmid DNA can be purified with a yield of up to 20μg from overnight cultures of up to 5mL LB, 3mL 2xYT, or 3mL TB. Using the NucleoSpin™ Multi-96 Plus Plasmid kit allows simultaneous processing of 96 samples within about 45min. The procedure is based on an alkaline lysis plasmid miniprep protocol. Bacteria are harvested by an initial centrifugation step. After resuspension of the pelleted bacteria and alkaline lysis the crude lysates are cleared by vacuum filtration with the NucleoSpin™ Plasmid filter plate. DNA is reversibly bound to the silice membrane of the NucleoSpin™ Plasmid Binding plate in a second filtration step. High purity of the final plasmid DNA preparation is achieved by removal of cellular compounds, salts, detergents, and other contaminants in subsequent washing steps. An additional washing step with buffer AW is recommended for E. coli host strains with high endogenous nuclease activity and for better performance of critical enzymatic reactions like long range DNA sequencing.
