Description
The Mouse Vascular Endothelial Cell Growth Factor Receptor 2 (VEGF R2/VEGFA) ELISA quantitates Ms VEGF R2 in mouse serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Ms VEGF R2. Principle of the method The Mouse VEGF R2 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Vascular endothelial growth factor (VEGF) is a major growth factor for endothelial cells. VEGF are a family of closely related growth factors having a conserved pattern of eight cysteine residues and sharing common VEGF receptors. VEGF Receptor 1 and VEGF Receptor 2 are both expressed in an endothelial cell-specific manner. They are detectable in virtually all tissues in adults and embryos. Monocytes express VEGF Receptor 1 and VEGF Receptor 2. VEGF Receptor 2 is involved in commitment of endothelial-cell lineages and to cell proliferation, while VEGF Receptor 1 seems to be responsible for guiding endothelial cells into the proper spatial organization of the lumen-containing vessels. VEGF Receptor 2 binds VEGF with high affinity but does not bind PIGF. The signalling and trafficking of this VEGF receptor are regulated by multiple factors, including Rab GTPase, P2Y purine nucleotide receptor, integrin alphaVbeta3, and T-cell protein tyrosine phosphatase. Mutations of VEGF are implicated in infantile capillary hemangiomas.Specifications
