The Human Cardiotrophin 1 ELISA quantitates Hu Cardiotrophin 1 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu Cardiotrophin 1. Principle of the method The Human Cardiotrophin 1 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Cardiotrophin-1 (CT-1) is a member of the IL-6 family of cytokines, which signal through gp130 receptor complexes. gp130 complexes with several different receptor subunits to transmit signals from Cardiotrophin-1, IL-6, LIF, OSM, CNTF and IL-11. Cardiotrophin-1 binds to and activates the leukemia inhibitory factor (LIF) receptor/gp130 receptor complex and has been shown to induce hypertrophy in cardiac myocytes in vitro. Cardiotrophin-1, a secreted protein expressed at high levels in myocardium during cardiogenesis, has been shown to promote proliferation and survival of embryonic cardiomyocytes, suggesting a role for Cardiotrophin-1 in the activation of gp130 during cardiac development.Specifications
