D-Cycloserine, CAS # 68-41-7, is a cyclic amino acid analog of D-alanine, originally isolated from Streptomyces species. It is synthetically formulated and exhibits antibiotic properties through its inhibition of bacterial cell wall synthesis.

General description

  • D-Cycloserine was originally isolated from Streptomyces orchidaceus and is currently formulated as an oral antibiotic
  • It is incompatible with strong oxidizing agents but stable under basic conditions, with the greatest stability at pH = 11.5
  • D-Cycloserine inhibits L-alanine racemase and D-alanine:D-alanine ligase, 2 enzymes responsible for bacterial cell wall synthesis
  • When ingested, D-Cycloserine traverses the blood-brain barrier, potentially causing neurological side effects such as, dizziness, nausea, depression, tremors, etc.

Applications

  • D-Cycloserine is a broad-spectrum antibiotic used as a second line agent for treatment of drug-resistant tuberculosis, always in combination with other antituberculosis agents
  • It is an excitatory amino acid and partial agonist of the NMDA receptor in the central nervous system (CNS), thus playing a potential role in the amelioration of neuropathic pain
  • D-Cycloserine has also been shown to play a role in neural connectivity, and is therefore used to study treatment applications in anxiety disorders and schizophrenia
