Description: The OX8 monoclonal antibody reacts with rat CD8 alpha. CD8 alpha is a member of the immunoglobulin superfamily which heterodimerizes with CD8 beta to form the CD8 antigen expressed on CD4+CD8+, double-positive thymocytes, the majority of MHC class I-restricted peripheral T cells, and on some macrophages. CD8 binds to MHC class I expressed on the surface of antigen-presenting cells during antigen presentation, and participates in T-cell receptor signal transduction through association with the kinase Lck. Applications Reported: This OX8 antibody has been reported for use in flow cytometric analysis. Applications Tested: This OX8 antibody has been tested by flow cytometric analysis of rat splenocytes. This can be used at less than or equal to 0.25 μg per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. Light sensitivity: This tandem dye is sensitive photo-induced oxidation. Please protect this vial and stained samples from light. Fixation: Samples can be stored in IC Fixation Buffer (cat. 00-8222) (100 μL cell sample + 100 μL IC Fixation Buffer) or 1-step Fix/Lyse Solution (cat.

00-5333) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation. Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically. Excitation: 488-561 nm; Emission: 775 nm; Laser: Blue Laser, Green Laser, Yellow-Green Laser. Filtration: 0.2 μm post-manufacturing filtered. CD8 (Cluster of Differentiation 8) is a cell surface glycoprotein found on most cytotoxic T lymphocytes that mediate efficient cell-cell interactions within the immune system. The CD8 antigen acts as a co-receptor with the T-cell receptor on the T lymphocyte to recognize antigens displayed by an antigen presenting cell (APC) in the context of class I MHC molecules. CD8 is found on a T cell subset of normal cytotoxic/suppressor cells which make up approximately 20-35 % of human peripheral blood lymphocytes. The CD8 antigen is also detected on natural killer (NK) cells, subpopulations of peripheral blood null cells, thymocytes and bone marrow cells. The CD8 co-receptor functions as either a homodimer composed of two alpha chains, or as a disulfide-linked heterodimer composed of one alpha and one beta chain. Both alpha and beta chains share significant homology to immunoglobulin variable light chains. The majority of CD8+ T cells express CD8 as a alpha/beta heterodimer. In HIV, the HIV-2 envelope glycoprotein binds CD8 alpha chain (but not the beta chain).
