Description: The A2F10 monoclonal antibody reacts with mouse CD135, also known as Flk2/Flt3. A member of the tyrosine kinase receptor family, this 135-150 kDa molecule is expressed by primitive progenitor cells in fetal liver and adult bone marrow. Two-color staining of adult mouse bone marrow cells with A2F10 and RA3-6B2 (CD45R/B220) or M1/70 (CD11b) reveals Flk-2-positive subpopulations of B cells or myeloid cells, respectively. Applications Reported: The A2F10 antibody has been reported for use in flow cytometric analysis. Applications Tested: The A2F10 antibody has been tested by flow cytometric analysis of mouse bone marrow cells. This can be used at less than or equal to 1 μg per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. Filtration: 0.2 μm post-manufacturing filtered.
FLT3 is a tyrosine protein kinase that is a member of the growth factor receptor tyrosine kinase family. The receptor is activated by binding of the fms-related tyrosine kinase 3 ligand to the extracellular domain, which induces homodimer formation in the plasma membrane leading to autophosphorylation of the receptor. The activated receptor kinase subsequently phosphorylates and activates multiple cytoplasmic effector molecules in pathways involved in apoptosis, proliferation, and differentiation of hematopoietic cells in bone marrow. Mutations that result in the constitutive activation of this receptor result in acute myeloid leukemia and acute lymphoblastic leukemia.Specifications
