The monoclonal antibody 1G5 recognizes human alpha catenin. Alpha catenin plays a role in cell-cell adhesion through its function in the binding and bundling of actin. The N-terminus of alpha catenin binds beta catenin and the C-terminus binds actin. Monomeric alpha catenin acts as a physical linker between the cadherin-beta catenin complex and the actin cytoskeleton and is stabilized by dimeric alpha catenin found in the cytoplasm. Alpha catenin also binds other actin-related proteins, including vinculin, alpha-actinin, ZO-1, afadin, formin-1, and Rho. Alpha catenin is absent in some tumor cell lines and reduced in some human carcinomas.
