Corning® NutriStem® hPSC XF Medium

Support hiPS and hES growth and expansion in a feeder-free environment with NutriStem hPSC XG Medium from Corning.

Enhance the single-cell culture of human induced pluripotent stem (hiPS) and human embryonic stem (hES) cells in a defined, xeno- and serum-free environment. Without the need for high levels of bFGF and other stimulatory growth factors or cytokines, low-protein NutriStem medium contains only the most essential components required for successful hiPSC and hESC maintenance, growth, and downstream differentiation.

Complete and ready-to-use, NutriStem provides a simplified medium for consistent performance, as well as increased reproducibility in hPSC long-term culture.

Superior Growth

NutriStem medium enables excellent proliferation of undifferentiated hES and hiPS cells. Proliferation of H1 hES cells cultured in Corning® Matrigel® Matrix-coated 96-well plates in NutriStem and the leading competing medium for feeder-free culture. Medium was changed and proliferation was assessed every 24 hours in culture.

Normal Colony Morphology

H1 hES cells (panel A) and ACS-1014 hiPS cells (panel B) cultured in Corning NutriStem hPSC XF Medium on Corning Matrigel matrix-coated plates display colony morphologies typical of normal feeder-free hES and hiPS cell cultures, including a uniform colony of tightly compacted cells and distinct colony edges.

Embryoid Body Formation

Embryoid bodies (EBs) were generated from H9.2 hES cells cultured for 16 passages in NutriStem hPSC XF Medium on Matrigel matrix as an evaluation of pluripotency. The pluripotent H9.2 cells were suspended in serum-supplemented medium, where they spontaneously formed EBs containing cells of embryonic germ layers. The following cell types were identified by examination of the histological sections of 14-day-old EBs stained with H&E: (A) neural rosette (ectoderm), (B) neural rosette stained with Tubulin, (C) primitive blood vessels (mesoderm), and (D) megakaryocytes (mesoderm).

Taratoma Formation

H9.2 hES cells were cultured for 11 passages in NutriStem hPSC XF Medium using a human foreskin fibroblast (HFF) feeder layer. The hES cells were subsequently injected into the hind leg muscle of SCID-beige mice for in vitro evaluation of pluripotency. The following tissues from all three germ layers were identified in H&E-stained histological sections of the teratoma 12 weeks post-injection: (A) cartilage (mesoderm), (B) epithelium (endoderm), and (C) neural rosette (ectoderm).

Extra cellular matrix

Corning® Matrigel® hESC-Qualified Matrix, *LDEV-Free

Corning Matrigel matrix is a solubilized basement membrane preparation extracted from the Engelbreth-Holm-Swarm (EHS) mouse sarcoma, a tumor rich in extracellular matrix proteins, including Laminin (a major component), Collagen IV, heparin sulfate proteoglycans, entactin/nidogen, and a number of growth factors.

Quality:

  • Mouse colonies routinely screened for pathogens via Mouse Antibody Production (MAP) testing
  • Extensive PCR testing is performed to screen for a number of pathogens
  • Tested and found negative for bacteria, fungi, and mycoplasma
  • Protein concentrations determined by Lowry method
  • Endotoxin units measured by Limulus Amoebocyte Lysate assay
  • Matrigel Matrix gel stability is tested for a period of 14 days at 37°C
  • Biological activity is determined for each lot using a neurite outgrowth assay.

Preparation and Storage:

Store at -20°C. Avoid multiple freeze-thaws.

Corning® rLaminin-521 (Human)

Support long term self-renewal of human pluroportent stem cells (hPSC) and neureal stem cells (NSCs)

Corning ® rLaminin-521 (Human) is an animal-component-free recombinant heterotrimer composed of a5, B2, and y1 chains expressed in a mammalian cell culture system. The Corning rLaminin-521 (Human) surface supports such key benefits as ROCK inhibitor independent, single-cell expansion of PSCs and inhibition of spontaneous differentiation, improving hPSC culture ease and efficiency.

A collaboration focused on bringing together better solutions for stem cell research

Corning has partnered with BioLamina for the supply of recombinant human laminin 521 (rLaminin-521), a next generation surface that enables feeder-free, single cell passaging of pluripotent stem cells (PSCs) without ROCK-inhibitor. Now we've gone one step further and are working together to combine BioLamina’s expertise in recombinant laminin expression with Corning’s extensive line of vessels and proprietary surface coating technologies. This partnership brings robust and scalable expansion solutions to the emerging stem cell processing market, enabling our customers to scale-up effectively and mitigate risk.

Now backed by Corning’s large network, worldwide distribution channels, and cell culture portfolio, researchers globally have better access and availability to rLaminin-521 (Human) than ever before.

rLaminin-521 (Human) joins the Corning ECM mimetic and advanced surfaces family: animal component-free surfaces suited for stem and progenitor cell expansion under cGMP processing environments.
